چکیده

facilitating distinction among the different types of aspartic proteinases. Thus, while considerable progress has been made in elucidating the structure, activity and importance (biological and commercial) of the aspartic proteinases, much remains to be learned about the distinctions in molecular architecture and how these are reflected in the functions of the various enzymes. Inhibitors (naturally occurring and synthetic) have permitted detailed biochemical and crystallographic investigations to be made but an understanding of the selectivity of such inhibitors may be of just as much importance for the design and synthesis of specific inhibitors for use therapeutically in controlling individual aspartic proteinases (e.g. renin). These and other aspects will be discussed in the articles which follow this introductory review.

Predictive Modeling of Phenylpiperazine Derivatives for Renin Inhibition.

The renin–angiotensin–aldosterone system is the well established endocrine system having significant role in preserving hemodynamic stability. Renin is secreted from the juxtaglomerular cells of the kidney. Phenylpiperazine derivatives have been reported as human renin inhibitor. To perform predictive QSAR modeling for 27 phenylpiperazine derivatives as renin enzyme inhibitors. The IC50 values ...

Renin Inhibitors in Foodstuffs: Structure-Function Relationship

Renin, a highly specific aspartic proteinase originated from kidney, is a ratelimiting enzyme in renin-angiotensin system. Recently, we developed effective expression system for recombinant human (rh) renin using baculovirus-insect cell expression system, and simple and rapid assay method for rh-renin using internally quenched fluorogenic (IQF) substrate. Using the purified rh-renin and the IQF...

Statine-containing dipeptide and tripeptide inhibitors of human renin.

Dipeptide and tripeptide derivatives containing a statine residue were synthesized as inhibitors of human renin. ES-305, bis[(1-naphthyl)methyl]acetyl(BNMA)-histidyl-statine 2(S)-methylbutylamide was found to be a highly potent inhibitor of human renin with a Ki value of 1.7 X 10(-9) M. Dipeptide derivatives with the BNMA group at the N-terminal (BNMA-Val-Sta-isoleucinol [ES-313], BNMA-Leu-Sta-...

Proteinase activity of human renin preparations: the International Reference Preparation (Renin Standard 68/356).

1. Several commonly used preparations of human renin, including the International Reference Preparation (Renin Standard 68/356), were examined for the presence of contaminating proteinase activity by using a 14C-glycinated bovine haemoglobin substrate assay. 2. All of the human renin preparations tested cleaved haemoglobin even in the presence o di-isopropylfluorophosphate and ethylenediaminete...

Highly potent and specific inhibitors of human renin.

Small peptide analogues representing the C-terminal portion of angiotensin I sequence were designed as inhibitors of human renin. Among synthesized compounds, benzyloxycarbonyl (-"Z")-(1-naphthyl)Ala-His-leucinal (ES-188), Z-(1-naphthyl)Ala-His-statine ethyl ester (ES-226), and Z-(1-naphthyl)Ala-His-statine 2-methylbutylamide (ES-254) markedly inhibited human and primate renins (inhibitory conc...

New modalities for treatment of diabetic nephropathy: a mini review

Background and aims: Diabetic nephropathy (DN) is the most common cause of end-stage renal failure which could increase the risk of cardiovascular disease and morbidity and mortality in patients. The aim of this study was to investigate new modalities for treatment of diabetic nephropathy. Methods:This study was a mini-review research to investigate drugs that are used for DN treatment. Resul...
